Al Shabab defeated Al Gharafa 1-0 in Riyadh.
The hosts left it as late as could be as a stoppage-time penalty from Nasser Al Shamrani won the game.
The Qatari team played for much of the game with ten men as Ahmed Fares was sent off in the first half.
With time running out for Al Shabab, Fisal bin Sultan was brought down by Saad Al Shammri and Nasser Al Shamrani made no mistake from the spot.
Persepolis also booked their place in round 2.
The Iranian champions saw their game against Sharjah cancelled as the UAE team withdrew from the competition.
Al Gharafa’s loss means Persepolis progress.
